We know he's already aboard during the early days of Kirk's first five-year mission in 2265 and is serving in the Sciences department as a staff physicist. It's always possible.

We don't know. We first see him in WNMHGB, but nothing about when he joined the crew

There's also Chekov, but I think at this point in the timeline, he's in high school.

He was 22 in Season 2 of TOS, making him fourteen in Season 2 of SNW. His Academy years are 2263-67.
We don't see Chekov until S2, though he logically had to be there during S1, in order for him and Khan to have met, though Chekov didn't recognize Harry Mudd in I Mudd. He therefore likely transferred aboard sometime between Mudd's Women and Space Seed.
