My favorite was Klag from TNG S2 episode "A Matter of Honor." Even though he got taken down by Riker, I still thought he came across as somewhat intimidating, while at the same time espousing the warrior-culture of the Klingons believably ("Look around you Commander, there are no old warriors.").
I really disliked most of the Klingons from the TOS films. Their definitive look was created during that time, but they never felt as gruff or intimidating as they did in TNG.
Kang from TOS was awesome. Kor wasn't a little too calm and talkative to be the best representation of a Klingon, but his lines and the way delivered them was magnificent.

Tough question. I guess for TOS, Kor. In the TOS films, Chang. Everything after, Worf.
Worf is the definitive Klingon for me (I grew up in the TNG era)
I would say the B'Lanna is second on the list (VOY touched on a LOT of Klingon stuff)
and Martok a close third.

This is a tough call. Part of me just wants to say Chang for sentimental reasons. The Undiscovered Country was among the first Star Trek stories I watched, and for years, Chang defined my idea of what makes Klingons awesome. I still credit him for the fact that Klingons are my favorite Trek species.
B'Elanna is my favorite character in Trek, and while she perhaps seems too human to be voted best Klingon, her resistance to being Klingon actually did more to define the species than a gruff Next Generation Klingon shouting about honor with no sense of what honor is ever could. It's kind of like how Spock's resistance to being human actually says a lot about humanity.
But the win has to go to Kor, for first establishing what Klingons are and, in my mind, what Klingons should be: cunning, sophisticated, and with a tinge of humor and good spirits to their brutality. Kor's debate with Kirk about what it means to be human is, for me, one of the franchise's defining moments. Long before Martok came along, it was Kor who appreciated the brutal, warrior potential of the human species. Also, I tend to dread original Star Trek/Generation-era crossovers, as I feel they often don't get the original characters right or give them satisfying stories ("Sarek," "Unification," Generations all come to mind). But Kor's stories in Deep Space Nine (particularly "Once More Unto the Breach") were every bit what he deserved.

I know I'm supposed to say Worf, because of my love of TNG and the show would not be the same without him. But he did too much to demystify Klingons and I did not care for most of it.

I don't want to hear anymore of that assholed shit, goddammit, stop with the Klingonese, already.

I feel like Christopher Lloyd's voice is too distinct to lose himself in the role. To me it was like Emmet Brown surgically altered to look like a Klingon, while acting really mean. Maybe if I had never seen Back to the Future I would feel differently.

I'm not really up on most of Christopher Lloyd's repertoire, to be honest. The only other thing I know him from is The Adams Family movies. The characterisations are so wildly different that I'm convinced that Christopher Lloyd's a good actor. When I see Kruge choking the shit out of that worm-monster, I'm like "...woe!!!" He's badassed, but he also has a sense of humour about himself. And I'm sure that when he tells his Men to feed his dog, he knows they're shitting a gold brick and it gives him a chuckle. The Klingonese in TSFS is tolerable because, for one thing, it's seldom used and for another ... you know it was new and different, then. Now, it's stale ... caricaturish ... lame.
