Some people feel that all the worldbuilding that has been done has constrained the situations for Star Trek somewhat, at least in the Alpha, Beta, Gamma, and Delta quadrants. Some people may or may not be right.
Screw it, we Andromeda now. New galaxy, new setting, and nothing holding writers back besides maybe picking up that probe destined for the Kelvans early on in the first season, possibly episode one. Stick a transwarp coil in an Enterprise and a few ships to help it along - Andromeda is a long way away, and not even a Big E can explore it all on her own - and if Dark Frontier is to be believed, you could get to Andromeda in a little less than a year.

Although even in "Errand of Mercy," which was the very first Klingon episode, the Organians predicted that the Federation and the Klingon Empire would one day be allies . . . .

I'd like this too, something about humanity adjusting to their new existence. I speculated a little about what it might be like hereI'd like a Star Trek show that paid attention to the logical implications of their plot-solving technology of the week.
Chemicals that can make people move faster than the blink of an eye, develop psychic powers, and tech that can de-age you at the touch of a button. These things would be just as epoch-making to the evolution of a civilization as faster-than-light travel was.
In short, don't ignore the McGuffins and Phlebotinum that you've decided DO exist in your canon.
