Up until about 100 years ago, there was one question that burned in every human, that made us study the stars and dream of traveling to them. Are we alone? Excellent choice.
When faced with this question, I am always inclined to say "Star Trek: First Contact". Captain Picard essentially defines what the Star Trek philosophy is to the character of Lilly. In addition, the end of the movie allows one to witness literally the birth of Star Trek. It doesn't hurt that the movie is perhaps the most accessible and interesting to a non-fan.
In terms of exlusively episodes, "Darmok", "The Devil in the Dark", "Demons"/"Terra Prime" and especially "The Measure of a Man" strike me as strong candidates.
