It's from the King Kong parody included in Treehous of Horror III, from season 4.
Randomly at work I was reminded of the "Not Lenny!" moments.
Homer: The coolant tank just blew and they're taking Lenny to the hospital.
Marge: Oh, no, not Lenny -- not Lenny!
Homer: Yes, I'm going to have to work late instead of seeing you and the kids, which is what I really want.
Marge: Okay, sure. Kids, turn off the TV. I have some bad news about Lenny.
Bart/Lisa: Not Lenny!
Later in the episode, they discover it's a hoax.
Marge: "Normally I'd be mad, but I'm just so glad Lenny is alright." (Looks over to a crochet she made for Lenny marked "Get Well Soon".)
Homer: "Hey, they are laughing at me. The first thing I'm gonna do when I get into work tomorrow is punch Lenny in the back of the head!"
Homer: Because People are stupid! That's why everybody does EVERYTHING!
I was having a bad day the first time I heard that, and that line made me laugh so hard, I shook right out of it.
The super quick shot that follows of Homer actually punching Lenny in the back of his head as he takes a gulp of coffee (forcing him to spit out the coffee) is one the biggest laughs I ever got from the show. Truly one of the greatest 'small' moments ever. Excellent choice, good sir.
"Your off the case McGonical."
"No, your off YOUR case chief."
"What hell is that supposed to mean!?!"
Homer: "THAT MEANS HE GETS RESULTS YOU STUPID CHIEF!"
Lisa: "Dad, sit down."
"Come along, Bort."
"Were you talking to me?"
"No. My son is also named Bort."
"Hello. I'm Mr. Snrub and I come from a place far away. Yes, that'll do."
Ah yes, the moderator anthem. We loaned it to the Stonecutters for that one.
Smithers: "I like the way SNRUB thinks!"
Homer: "Hello, my name is Mr. Burns. I believe you have a package for me."
Postal Worker: "Okay, Mr. Burns... What's your first name?"
Homer: "...I DON'T know."

Homer: "Look at Kent Brockman, thinks he's so great just because he has so much money, but there are some things money can't buy."

I love Homer finding out he knew his middle name all along with out realising it, which is why I combine two of my favourite bits right here.

"Do it for her" - gets me every time
Lisa gives Milhouse a note, asking him to pass it to Nelson in class. In class, Milhouse does so, Nelson reads it "someone likes you." Cut to paramedics rushing Milhouse to an ambulance.
Also, from the same episode, Lisa and Nelson kiss. Jimbo or Kearney or whoever sees this. "Dude, you kissed a girl. That is so gay."
Martin (as Lizzy Borden): Forty whacks with a wet noodle, Bart!
Bart: Dad, why did you bring me to a gay steel mill?
