I don't know if it's ever been spelled out directly, but I've always imagined Vaughn having a little bit of a midwestern Indiana twang in his voice. Just a touch, but definitely part of the flavor. Like Hal Holbrook or maybe Sam Waterston.

Assuming Vaughn is in good enough health, I doubt he looks anywhere even close to his true age. If people routinely live that long, probably everyone looks younger than they are. (For example, Picard is in his 70's now, but doesn't look it.)
