From "The Doomsday Machine":

"Then you mean you're the lunatic who's responsible for almost destroying my ship?" (To Commodore Decker).

From "The Changeling":

"You are flawed and imperfect! Execute your prime function!" (Convincing Nomad to destroy itself.)

From "Mirror, Mirror":

"It is useless to resist us!" (To the Halkans, and perhaps, the inspiration for the Borg's catchphrase, "Resistance if futile.")

(I realize that's more of an "exchange" than a "one-liner", but I always liked it because Kirk set up McCoy in the BEST way!) :)

Yeah, it actually goes more like this...

KIRK: Bones, there's a...thing out there--
BONES: Why is any object we don't understand always called a "thing"?
KIRK: --headed this way. I need you. (beat) Dammit, Bones! I need you...badly!
BONES: Permission to come aboard?
RAND: (filtered) Permission granted, sir!
BONES: Well, Jim, I hear Chapel's an M.D. now. I'm gonna need a top nurse, not a doctor who'll argue every little diagnoses with me. And they probably redesigned the whole sickbay, too! I know engineers, they love to change things.
 
The one line I remember because it showed how the show was about more than just space monsters,
From The Man Trap:
"Just thinking about the buffalo."


And from The Ultimate Computer, Kirk quoting from the poem "Sea Fever," "All I ask is a tall ship, and a star to steer her by." and I can't remember the rest, but when he relates that to being a starship commander. About how the stars are still there, and the ship. Someone must remember those exact words.

Hmm, just thinking, but, I doubt he ad-libbed that. He was saying the same thing the tribble salesman said earlier.
