If you can turn a TV show into a comic series...

LOST

It could decide to give some sort of far-ranging story that gave the basic purpose/origin of the island.
 
Firefly / Serenity. The occasional mini-series that Dark Horse's Scott Allie hopes for, and the "fill-in-the-blanks" mini-series we've already had set in between the series and movie, simply aren't enough for me. Since we'll never get a new television series flying, I'd love to have a new series exploring how the revelations in Serenity will affect the 'verse and the roles that Mal, Zoe, etc. will play in events. I like to imagine a new Browncoat uprising...

Failing that, I'd like to echo Theonethatis and say Crusade. Perhaps it could start with some of the finished-but-unproduced scripts. I'd want it to, like JMS originally intended, follow the "find a cure" plotline for a year or two, then move onto to the lingering Shadow influence within EarthGov and Excalibur going rogue. I've enjoyed JMS's original comics work far more than what I've read of his contributions to licensed superhero material, so it really seems only natural -- and a big-time missed opportunity -- for Crusade not to have a comics continuation.

I think it could be a fun read if written well.
 
LOST

LOST could actually open up some pretty interesting comic book stories. You could have some deeper exploration into the Island's past, but I honestly think I'd like to see more of the future of our favorite characters. We could see the future of the Island with Hugo and Ben, find out the circumstances that led to Hugo's inevitable death and how he found a replacement for himself.

Hell, we could have a whole series about Cindy and what happened to make her an Other!
 
You could do all kinds of stuff with Lost. Instead of an on going monthly comic I'd like to see it done in the miniseries format. You could do one for Dharma. Rousseau's time oin the island post Alex snatching and pre-crash, Widmore's attempts to find the island, secveral mini series about different people who went to the island over the millennia, etc.
 
I'm going to repeat an article from an old Wizard that I agreed with...their concept for a "24" comic was really interesting and I could get behind that. I think the show's formula of internal political struggle combined with external terrorist threat would work well for a comic. I'm surprised no one has mentioned "Heroes". I'd relaunch the series fresh as a comic.
 
To get more storytelling from something that's cancelled or ended. It's not exactly a novel idea. It worked for Buffy, Farscape, and several others.

I wouldn't mind seeing some more X-Files comics, some Lost ones, something in the the B5 universe. Lots of good stuff ripe for being comics out there.
