Best Line in all of Trekdom

"The truth is usually just an excuse for a lack of imagination."
-- Elim Garak

"It's mostly sugar."

-- T'Pol

"Five card stud, nothing wild, and the sky's the limit!"
-- Jean Luc Picard

"What does God need with a Starship?"
-- James Tiberius Kirk

from Call to Arms, after Sisko tells Weyoun he will not allow any more Dominion ships to come through the wormhole, Weyoun's response:

"You will not allow"

the sheer contempt in Weyoun's voice was fantastic - the very idea that someone would try and stop the Dominion doing what it liked.

"Well, that's the second time Man has been thrown out of Paradise."
"No, Bones, this time we walked out on our own. Maybe we weren't meant for paradise. Maybe we were meant to fight, struggle, claw our way up - scratch for every inch of the way."

"In every revolution, there's one man with a vision."

"They invade our space and we fall back. They assimilate entire worlds, and we fall back. The line must be drawn here! This far, no further! And I will make them pay for what they have done!"

And my personal favorite:
"The more complex the mind, the greater the need for the simplicity of play."

Here's another one:

Life-forms....
you tiny little life-forms....

you
precious little life-forms....
where are you....?

"Space... The final frontier. These are the voyages of the starship Enterprise, it's continuing mission:"
"To explore strange new worlds, to seek out new life and new civilizations,"
"To boldly go where no one has gone before."
- Picard Kirk and Archer.

It's not a single line, but I love this scene. It sums up the outsider attitude toward the Federation and the much of the real world's attitude toward America:

QUARK
And the worst part is, my only
hope for salvation... is the
Federation.

GARAK
I know precisely how you feel.

Quark suddenly gets an idea.

QUARK
Here, I want you to try something
for me.

Quark pulls out a glass and fulls it with a foamy brown
liquid.

QUARK
Take a sip of this.

Garak looks skeptically at the drink.

GARAK
What is it?

QUARK
A human drink. It's called root
beer.

GARAK
(eyes it suspiciously)
I don't know...

QUARK
Go ahead. Aren't you just a
little bit curious?

Garak hesitates a beat, but then takes a sip. He
immediately makes a face.

QUARK
What do you think?

GARAK
It's vile.

Quark is glad that someone agrees with him.

QUARK
I know.
(searching for bad things to say)
It's so bubbly and cloying and
happy.

GARAK
Just like the Federation.

QUARK
But you know what's really
frightening? If you drink enough
of it, you start to like it.

GARAK
It's insidious.

QUARK
Just like the Federation.

There's a long beat as the two aliens share their
common bond.

GARAK
Do you think they'll be able to
save us?

QUARK
(defeated)
I hope so.

Quark takes a sip of the root beer, then shudders.

(Pointing to a swastika banner.) "Look at our destiny. The field of red; the purity of German blood. The blazing white circle of the Sun that sanctified that blood. No-one can deny us; no power on Earth or beyond. Not the Christian savior, not the God of the Jews. We are driven by the very force that gives life to the Universe itself."
