I think my favorite line was when Cupcake said to Kirk at the bar that there was four of them. Kirk's response, "Well, go get a few more and then we'll have a fair fight," so sumpin or other. Awesome.
I also thoroughly enjoyed Scotty's lines. All of them.

I think the humor is a large part of what made it such an enjoyable film and accessible for so many people.
I enjoyed most of the jokes (especially "Out of the chair" and "I have no comment on the matter"), but "Numb tongue?" made my eye twitch a bit. My mind jumped to Scooby Doo when Kirk said that and tried to give Uhura the message.
To make it clear I am not trying to be cynical, I find myself laughing often at TOS. I love how a previous poster described it as "happy humor"... I completely agree. It's silly, light-hearted, and wholesome. I just want to wrap it all in a big hug and squeeze the stuffin' out of it.

i liked how they intertwined the humor so it relieved tension in otherwhise a pretty serious film.

even the ultra serious city had touches of humor thorugh it.
as for scotty well try the end of by any other name or tribbles as to how he wasnt played totally seriously all the time.

It was kind of obvious, as a gag, but I liked the way Yelchin played it.
Yes I did...I thought it was a cute scene. I was afraid of not liking Chekov's accent at all because I heard some negative things about it beforehand.....but I actually loved it! Loved it so much that I've tried desperately (alright not desperately) to get the Chekov toy from BK because he says his name which would of been a hoot. But it's not meant to be....
I think the moment that cracks me up the most is the Red Shirt's excitement about "kicking some Romulan arse" or whatever, followed by a blank stare on the part of Sulu and Kirk.
It's the kind of thing that's satisfying as a long time fan because really the longer you've been following Trek the funnier that moment is.

I have always appreciated the humor of TOS.
I was watching The City on the Edge of Forever last night, and even though it dealt with the death of a storyline character, there were plenty of instances of laughable interaction with a lot of characters. However, the usual last scene between the characters and having a light hearted moment was not warranted and stay in 'character' to the episode.
I will take humour in any episode as long as it is done right and with purpose. Just like there aren't all serious days at a job, there are serious days in TOS (Balance of Terror) to non-serious days (I, Mudd).
I liked when Chekov was running to the transporter room mumbling "I can do that, I can do that!" No offense to Wil Wheaton, but that's what a precocious wunkerkind should act like.

Thank you. I'd forgotten that.Yes. I am foreign and sometimes I find English to be a mouthful and I found it funny that the computer wouldn't recognize the words. I also found Chekov's mix of youthful enthusiasm and brilliance made him a very enjoyable, funny character with too few scenes.
When I went to the first viewing on opening night, people laughed at every joke. The second time around... not a one. Maybe it was just because everybody had already seen it?
There are different dynamics with different audiences. People have to start laughing for others to follow. The French audience I was a part of laughed very little. At this kind of movie, it's always the same: I feel as if I am almost the only one. I knew it would be like that, but... it was a 3/4 full, huge theatre!
I guffawed a little louder than most 
but I liked the humour. I enjoyed Simon Pegg's lines and look forward to what he has to do in the next flick. My favorite moments were when Kirk was having the allergic reactions. McCoy keeps injecting him: "THHHstop that!"
