I'd like another episodic plus occasional mini-arcs show in the vein of TNG where we jump forward to the mid-to-late-25th century aboard a new iteration of the Enterprise (H, I or K, not J) on a mission to explore the most distant reaches of the Milky Way and its nearby satellite galaxies.
The traditional enemies are allies now or non-threats (Cardassians, Romulans, Klingons, Xindi and Ferengi routinely serve aboard ship) and there are new beligerents, competitors and potential friends on the distant frontiers of the galaxy to meet and contend with.
The ship is now capable of traversing the galaxy in a matter of months rather than decades, and can even reach some of the closer minor satellite galaxies without great difficulty. We can sometimes revisit species encountered by Voyager and see how they're doing a century later.
Both the Bajoran wormhole and the now artificially stabilized Barzan wormhole (and others discovered since) also provide shortcuts back to the Federation Alpha/Beta core worlds from the Gamma and Delta quadrants should a faster return be nescessary.
