Joel, dude, it sounds like you're planning your book as it it were a paint-by-numbers kit. Nobody here can tell you where the colors go. Either you know how to write or you don't. Just do it, and--most importantly--learn from the result.

Basically just go with whatever feels right for the story, and the context of the story. If something just doesn't mesh well, or doesn't feel right, you know it's best to drop it, and leave it out.

Helpful for those of us who do text messaging every now and then; although, I have to remember to not use 'text/forum speak' when I type business emails ....

I find a paraphrasing from Alice in Wonderland to be apt.

Start writing at the beginning and when you get to end, revise. Then stop.

*I know some publishers are nitpicky; so, would a prologue count as one of my three chapters that I would submit in my proposal?

^As has been stated before, the sample chapters should be the first three chapters of your book. You need to prove that you're able to write a strong beginning, one that can grab the reader and keep them interested. So yes, you start at the beginning, regardless of whether it's called "Prologue" or "Chapter 1."
