Not quite. The generation 1 name was "Twilight Twinkle," but Hasbro lost the rights to it some years back. When asked on DeviantArt if the new name was a reference to the Twilight series, Lauren Faust answered only "It passed legal." Make of that what you will.

(I do think giving that name to the nerdy book-lover with no interest in romance was a deliberate tweak at the movies and novels, though.)

Well, the most Trekkish episode is the two-parter that starts off season 2 called "The Return of Harmony". It features John DeLancie voicing a Q-like character. You should probably watch an episode or two set before that first to get a feel for the characters and the show, maybe "Look Before You Sleep" and "Bridle Gossip" which give insight into the main characters.

Sorry that I have to make a new post to reply, but if I edited my previous one, all the youtube links would disappear. Anyhoo, as to my answer, I still don't think it's a reference to Twilight, but even so, Twilight Sparkle is a much better use of the name.

The pilot's important. It lays the groundwork for all of the main character ponies, which is why I showed more than three.

One of the things that brought me to love the show is the near lack of cynicism. The idea that people, er, ponies are innately good and that you can depend on your friends when you need them the most.

i tried, i really tried, to watch that episode with De Lancie in and i watched about a minute before i had to stop. i couldn't abide the sickening cuteness any longer.

I get why other people seem to like it (colourful, optimistic, quirky humour), but I don't. Not really. I mean, it would be tolerable if I had a little kid who watched it, but I don't enjoy the show to the same degree as I have a lot of other animated programs that are aimed at kids but have adult content on another level.

I watched the pilot and the following six episodes. Was occasionally amused, it's a high quality program as far as animation and voice goes, but I just don't get why some 20 or 30-something men seem to be all over it like it's the best thing going. I chuckled occasionally, had no desire to watch more once I checked it out. My wife was bored to tears for most of it.
