That would actually be awesome. A ancient, mysterious sci officer that's always angry and bitter because he lost his key to the whole universe. He has an odd affinity for bow ties too.
Taken the idea on a path that's less ripped off, the idea of an immortal being (without cliche powers and agenda) could be interesting. I don't think anyone has really done the concept justice (though beheadings are and will always be cool...err...so long as Al Qaeda or the band Queen isn't involved)
Federation ship: 2 Humans, Andorian, Vulcan, Trill, Bolian, Caitian
frequent ally ship: Klingons, Romulans, Gorn (not necessarily on the same ship), Hydran (with British translated voices)
adversaries: Interstellar Concordium, Kzinti/Mirak, Lyrans, Cardassians, Organians

Lots and lots of Klingons.
Humans ... sure, got to have them,
Andorian ... yes, plenty more to learn about them.
Vulcan ... maybe, we have the Vulcans fairly well figured out by now. Perhaps a reoccurring character.
Trill ...... no, other than the slug in the gut, what do they really they bring new to the table?
Bolian ... great, they're largely unknown, how about two who don't like each other?,
Caitian ... depend on if they can get the costume right. An interesting species and a reference back to TAS.
frequent ally ship:
Klingons ... like the Vulcans old news, replace them with a fresh young species from the planet "hip-hop party girls." Romulans ... been done to death, boring and used up ... next.
Gorn ... yes yes, endless possibilities. Use either FX or a practical costume. Mouth might be a problem.
Hydran ... I'm unfamiliar with them, which is good.
(with British translated voices) ... aliens who don't speak with a American mid-Atlantic accent? Are you mad?

With the exception of the Cardassians, sounds good, especially the Kzinti who always sounded so mean and powerful in Niven novels.
The Conn Officer is the Helm-Nav Officer.
I was actually reading a short-lived comic book series called Negation the other day--a sort of Firefly meets Star Wars sort of thing. Long story short, among the list of Human and non-Human characters was a "sexy lizard lady" who was sort of the group's heavy hitter. Perhaps she could be a member of a new alien race we've never encountered before...
I am currently working on a fanfic series (as announced in my sig), which is basically how I'd do a new Trek show. The crew consists of:
Captain, joined Trill, white male
Executive & Science Officer, half-Vulcan half-Romulan, white male
Helm, human, dark (as in Indian) female
Doc, human, black male
Security, Andorian, blue male
Chief Engineer, human, white male
generalist Ensign, Deltan, white female

A Vulcan\Romulan hybrid-finally follow through on failed ideas for Savaak (orig script) and T'Pol (proposed idea).

Xindi (Arboreals were my favorite)

A Gorn.
I guess you need a self-aware hologram, too.

The Trill is a must, they didn't do enough with Jadzia and Ezri.
The Betazoid can not be like Deanna Troi or Lwaxana forthat matter, so preferably male.
Romulan, I think in JJverse the Federation just might be allied with the Romulans rather than the Klingons.
Andorians are just cool looking and it would be nice to see them.
Orion, maybe a human/Orion hybrid female so the pheromones weren't as strong as either the counselor or science officer.
And Ktarians, have never really been used much.
