I have been through a similar experience with my wife. She also had no interest in sci-fi in general nor in ST specifically. I found that the best way to introduce her to ST (with the ultimate goal of making her a fan) was to focus on episodes that are accessible to as wide an audience as possible (and not just to trekkies/trekkers/sci fi geeks, etc...). Here is a my list of suggestions with a brief explanation as to why they work best IMHO for this type of situation:
The Trouble with Tribbles: It's cute and funny and you don't need a lot of ST background to get what is going on.
Wolf in the Fold: A whodunit dressed up as Star Trek. It's dramatic and suspenseful and introduces very subtly the level of tech in the ST universe.
Amok Time: After my wife saw this, she became very interested in the character of Spock because this episode is more about relationships and of how loyalty can be strained when our professional and personal lives collide.
Charlie X: Charlie is a fish out of water and that makes him very easy to relate to for a newbie.
The Squire of Gothos: A colorful character who provides a nice balance of terror and humour and who leaves you with an interesting topic of discussion at the end.
City on the Edge of Forever: An excellent story...period!
The Ultimate Computer: Despite the occasional tech-talk from Daystrom, the basic conceit of the story is very accessible even to the newbiest of newbies.
Once she has attained a sense of liking the show, then you might want to try to move onto the episodes that require a little more suspension of disbelief and forgiveness of cheap special effects. (Eg., Arena, Spectre of the Gun, Space Seed, And the Children Shall Lead--ahem!).
