I'm afraid I don't get the "poke" reference. Is it sexual? Or is it something to do with Facebook or Twitter (neither of which I use)?

It's a facebook thing. You can "poke" your friends and they can poke back, it's usually a way of getting their attention.

Kira: Wow, my shoulders were huge in the old uniform.

Nobody was really sure how nude pictures of Bashir got into the Defiant's system.
