Pardon if this has been covered in another thread, but I don't get in here as often as I used to...it sounds like he has one due out in latter 2012, but I don't recollect hearing if it's all-new post-Destiny, or Vanguard, or Mirror Universe, or something completely different. Can anyone share?

It's post-Destiny TNG.
I have several pet theories and rank speculation as to what would be covered, but Mr. Mack has not posted a spoiler yet.
I hope we might here something at the beginning of December as the publisher seems to have been in the habit of posting back-cover spoilers a year in advance and this trilogy is slated for December 2012.

I'm pretty excited to read more post-Destiny novels from David. Should be quite good, as should David R. George III's new books coming out.

Perhaps it can explain things that made no sense in that, like how Picard became ambassador, and when B-4 got Data's memory programmed into his own.

Yes, Mack writing a new trilogy should be epic.
As for Picard being an ambassador in Countdown. He's always been more or a talker than a fighter (although the latter he'll do if need be) and as he's refused several promotions to the Admiralty over the years, becoming an ambassador would be a logical move if he ever decides to stop commanding the Enterprise and doesn't want to retire.
That happened in Nemesis. Data downloaded his memory engrams into B-4 in an attempt to jump-start his intellectual development. It failed, but at the very end of the film, B-4 sang a snippet of the song Data had performed at Riker & Troi's wedding, suggesting that he might be able to access the memories to some extent after all. Countdown's writers simply went with the interpretation that the memories were eventually "awakened" and effectively turned B-4 into Data.
....which was depicted in the Star Trek Online novel "The Needs of the Many". But circumstances are different enough in the Destiny novels that an alternate resurrection will be needed should the Destiny books tie in to "Countdown"
Even though I actually enjoyed Countdown, I don't think I'd want to see the books be tied to it. Since the books have changed so much of the status quo since Nemesis, I'd rather see the books be allowed to continue to follow their own path rather than be forced into following CD.
