I could never see the Ferengi as villians but the role they play as being devious pains in the rear to Star Fleet and any others that cross their bank balances is brilliant.
Could the Ferengi have worked as the main villains?

Could they have worked? The short answer is no. After their botched introduction in The Last Outpost, they were pretty much set down the path that they eventually went. Fortunately, by DS9, the writers got them figured out, so they ended up working out pretty well.
Problem with the Ferengi is that they look a comical bunch almost like short freaky clowns. So, even their appearances do not make them a serious type of adversay.
