Eka Darville has confirmed that JJ's friend Malcolm will appear.
Breaking news! Sigourney Weaver has been cast as the unnamed villain in the Defenders.

I was going to crack a Ripley Hunter joke and then realized wrong universe.
The best guesses I've seen so far are that she's either a wildly different take on Typhoid Mary (which seems unlikely, but would undoubtedly be worth watching), or more likely, that she'll be the physical manifestation of Mephisto or The Beast (the demonic entity The Hand worship) in some form of adaptation of the Shadowland crossover from a few years back.
And if they're right, that begs the question, "Why look like that?" Which has in turn led to a couple of people I've read speculate that perhaps that's what Matt's mom looks like. Which would be a neat way to work in the dear Sister.
Though at this point, that's working under the assumption that she's a) an existing character and b) coming from the Daredevil side of things. Which could all turn out to be nothing. With no actual information, it's all fun and games with the guessing for now. Though I admit, having now read the Mephisto guess, I'm going to be just a wee bit disappointed if that one doesn't turn out to be true.
There was an Iron Fist clip shown at NYCC that probably has more relevance to Defenders.
In it, Danny mentions that his mission is to stop The Hand and that, "I'm the only one who can do it".

I seriously doubt it. They teased it when she was shot in the arm and without proper treatment by Luke and Claire might have lost it (paving the way for the cybernetic arm) but 1-2 episodes later (barely a day later in show time) she was moving about and handling herself as if it were just a scratch and minor nuisance and not a life threatening injury.

The second. In 2012, she starred in a Greg Berlanti-produced USA Network miniseries called Political Animals. But that's it, unless you count her two Saturday Night Live hosting gigs 24 years apart.
There is no Dana, only Zuul.
And if she is in fact a demonic entity, and they don't reference this line, then the writers should be ashamed of themselves.
Given that in Luke Cage, Shades really did ask Willis what he was talking about, a Zuul reference wouldn't surprise me in the least.
