Culber was (imo) the best candidate,
Some of the characters were likely off limits (too important to the story), of those not off limits he was the most likely to discover Ash's deep dark secret and "require" killing.
Culber's murder was obvious coming once Ash obviously was a Klingon (somehow) in disguise.Long before that scene.
His murder gave Stamets a personal tragedy. Another reason Culber's death at Ash's hands was telegraphed. Stamets was on the off-limits list.I thought he would be off limits since he was married and part of the very first gay romance of the very first gay character in Trek
