The Greatest Human Invention?

Genetic Engineering. I mean, the ability to completely change what a living being is on the most fundamental, previously inviolate level... that's pretty frickin' amazing.
 
The french press. Did you know you can make coffee AND foam milk with it? Amazing.
 
I'd go for the wheel and the lever, two things that have brought advancement in every field, including agriculture, industry, commerce, construction, etc.
 
If were talking about discovered/mastered then fire, as for invented it would have to be the wheel, no doubt.
 
For the single greatest invention for humanity, it will have to be the written language. Written language allowed humanity to begin to accumulate knowledge and to pass it on to future generations. Without the written language, we would've never been able to build on the discoveries of those that came before.
