Phew, that's a pretty general question. There have been dozens upon dozens of ST video games. I would recommend checking out the individual titles using a list like http://en.wikipedia.org/wiki/Star_Trek_games

In short, most Star Trek games for PC can be played with a conventional keyboard & mouse setup and do not require very short response times (excluding FPS titles like Voyager Elite Force of course).
