Give me a Connie (originally Starship Class) and I could rule the galaxy.
It's just a fact that it works more efficiently. No stopping to run Diagnostic X, Y or Z. No endless reconfigurations of the sensor dish to microwave Borg nachos. No releasing of gas du jour from the nacelles.
Connies get the job DONE, first try!

Ambassador Class, or if it a newish ship perhaps a Nebula, I doubt they would have a first command to a galaxy or other pride of the fleet.

Prometheus-class. Fast, new, and powerful. Someone also mentioned the Galaxy is like two ships for the price of one. Well, the Prometheus is three.
But I'd delete the EMH Mk. II because I don't like his personality. I'd replace him with a Mk. I. And believe it or not, I like the Mk. I's bedside manner.

First choice: Constitution Class refit just fo the sake of the status
Second choice: Defiant Class seems snug, cozy and a hell of a kick ass vessel
Third choice: Ambassador Class, little seen and under rated

For my first comand, I'd take something like the Miranda class, in the 24th century. I've always liked the design. It would still feel like a ship instead of a hotel. Being an older ship, perhaps it could be assigned to Starfleet Academy as a trainer, or something. I'd still get to teach, but I'd likely be able to avoid some of the really dangerous situations. Stay closer to home, but still get my space legs.

There are four classes I would be happy in the centre seat of: Sabre, Miranda, Excelsior, Ambassador.
Sabre - Small, agile and nippy
Miranda - The Ford truck of starships, what's not to like?
Excelsior - She may the "elder statesman" of the fleet, but its a classic
Ambassador - Big and comfy without being overly flashy like the Galaxy or Sovereign (a woefully under-used ship IMO)
