If the Romulans at long last are getting the development that Trek owes them, then let's keep up the momentum. The tension between them and the Vulcans and how all the Feddies get dragged into that fight needs to be explored. And no, I dunno how to reconcile it gracefully with canon. Tell a good story and I won't care!
All the other favorite ideas of mine are in the past (Birth of the Federation) or the future (revive and un-screw-up the Borg; tell us what happened after the Dominion War).
One thing they could do with the 23rd C is to bring back the Harry Mudd/Cyrano Jones style space rogue. I liked ENT's introduction of the Boomers as humans who had settled space and were outside the sanitized bounds of Earth society. How about more humans like that? I'm tired of humans always being Trek's most boring "aliens."
...but seriously folks. No 24th-C elements like Borg, Ferengi, Cardies or Dominion. Let's allow the different times to have their own character and not just make everyone an indistinguishable gray mass of Trek.
