Not too spoilery. Interesting little exchange, I presume from "Part II" (because the whole thing is in the print issue of TV Guide, but why wait?)

Kimmel: Is everyone on the island from the planet Earth?

Lindelof and Cuse: [Long pause] Yes.

Cuse: That may be one of the best Lost questions we've ever been asked.

Kimmel: Thank you.

Lindelof: When you get asked questions like that, you have to be very careful how you answer.

Nah, here's what it is: a chunk of the Solaris planet broke off and smashed into the South Pacific eons ago. An island grew around it, but at the center of the island is an amorphous mound of alien intellect, so alien that it lies beyond our capacity to comprehend, but still capable of whomping up some killer illusions, smoke monsters, immorality and other fun stuff.

So, that's what the long pause was about. Nobody on the island is an alien.
 
They're lucky Edward James Olmos isn't playing Jack. He'll walk at the first sign of aliens.
