The title of this thread is a line that Bones says in The Voyage Home. Even more than "Dammit Jim" and "He's Dead, Jim" I think this line encapsulates his character and his role on the show nicely.

So,, for the major characters, find a line of dialogue that you feel does the same. If you can think of a better one for Bones, by all means.
 
For Scotty, “Any man who could manage such a feat, I wouldna dare disappoint. She’ll launch on time, sir. And she’ll be ready.”

For Chekov, does a wordless scream count as a line of dialog?
 
[When asked what the logical purpose of his medals is]
Data: I do not know, sir. I suppose none, I just wanted them. Is that vanity?

I always thought that line summed Data up pretty well. You get the naivety, but also that great interest in himself. He seems slightly exhilarated by the idea that he might have performed an act of vanity, but the fact that he feels interest in it kind of defeats its own purpose, and hes back where he started again. Its moments like this that really give you a glimpse into his character, although I suppose it doesnt really sum up his role on the show.
 
For Scotty - "Now, drink your drink."

For Pulaski - "It does know how to do these things, doesn't it?"

For Worf - "On the Enterprise, I was considered quite humorous."

For Tuvok - "Shall I flog them as well?"

For Mayweather - "Aye sir, full impulse."

Spock: "I have been ... and always shall be ... your friend. Live long ... and prosper."
(Alternatively: "THE WOMEN!")

Picard: "The first time any man's freedom is trodden on we're all damaged. I fear that today..."

Sisko: "So... I lied. I cheated. I bribed men to cover up the crimes of other men. I am an accessory to murder. But most damning of all... I think I can live with it... And if I had to do it all over again... I would."

Good for the more mellowed-out movie Spock.

For TOS Spock:
“Being split in two halves is no theory with me, Doctor. I have a human half, you see, as well as an alien half, submerged, constantly at war with each other. Personal experience, Doctor. I survive it because my intelligence wins over both, makes them live together.”

McCoy:
“This is a big ship. I'm just a country doctor.”

Scotty:
“They called the Enterprise a garbage scow, sir!”
 
T'Pol: "You presume that my time with Humans has left me susceptible to their emotions."

"Humans believe that sometimes... you have to follow your instincts. Very illogical approach, but one I've come to embrace."

Think they encapsulate her experience on ENT...
