"Fire" is great too, and probably would have had more impact if I had seen the episode as it aired (I didn't get into TNG until the 6th-7th seasons).

I wouldn't say it's the best moment ever, but a contender that hasn't been mentioned: the opening of "The Child", with its triumphant fanfare, beautiful effects shots, and sweeping shot of the bridge, revealing Worf and Wesley's new uniforms and Riker's beard. It was not only the first episode of a second season that people might have wagered against a year earlier, but also long-delayed by the writers' strike. The show was back to stay, and this opening celebrated the moment.
I remember seeing that and just yelling "WHAT THE FUCK?!" out loud 
One of my favorite TNG lines is Q to Worf. "Ah Mr. Worf, eat any good books lately?"
"Fire" from BoBW was a great scene but the second part kinda kills it when nothing really happens. Although I think this is the first time we ever see anything actually coming from the deflector dish.

One of my favorite comedic bits is from "Captain's Holiday". When Picard finds out what the Horga'hn is for, his reaction is priceless.

The cliff-hanger ending of "Best of the both worlds" is chilling. The different reactions of Shelby and Riker to the news that Picard is a Borg are interesting. Especially when you look at Shelby's speech earlier, when she accuses Riker of not being able to make the big decisions and sit in "the big chair."

I'd also add, for sentimental reasons, Riker's earlier speech, as he demolishes the notion of Data's sentience, finally turning him off. 'Pinocchio is silent, his strings have been cut.' It was the first moment of ST I'd ever seen. I guess it's why I'm here today.

It may not feature a member of the main cast (for this particular incarnation of Star Trek) but O'Brien's line from "The Wounded" has always been one of my favorites: "It's not you I hate, Cardassian. I hate what I became because of you."

Another good moment is when Worf kills Duras. Whenever I watch that I scream YES!! out loud when Duras gets it.
