I don't see why she would "let" him get close for any reason. Her knowledge of the neck pinch is immaterial.
 
Okay he pretends to stumble and she moves in for the kill, he twists, leaps and pinches.

(That's a little to acrobatic for Spock, LOL)
 
Isn't Buffy, supernaturally augmented? She is a "slayer" after all, if I remember the series correctly (been a long time).

Spock wouldn't stand a chance, unless he was somehow able to clamp on a mind meld.
 
Gotta say that someone who is trained and born to kill vampires and demons,can't be beaten by a vulcan,a klingon or any other science fiction aliens.
 
Surely, Spock would find fighting Buffy highly illogical and instead rig up a solar gun to help her dispatch vamps without all the fisticuffs.
 
I think some random throwaway line on DS9 established the average Vulcan's strength as being three times that of a human. 'course, Spock is only half Vulcan, and feats of strength were never really his specialty.

Buffy's faster, too.

Spock versus one of the vampires would be a more even, and thus more interesting match. His nerve pinch advantage might not work in that case, too...
