Every Star Trek series has had its share of jokes and lightheartedness. Which one did it best?

For me, it's DS9 hands down. Despite being the darkest show, it has the best selection of lighthearted episodes like "In The Cards", "Take Me Out To The Holosuite" and "Trials and Tribble-Ations", and even outside of those, the large and diverse recurring cast and the way the regular characters interacted meant that there was always ample opportunity for some witty dialogue or some Ferengi farce. The Bashir/O'Brien bromance was my favourite pairing out of all the series.

I just wish they had managed to rekindle the Worf one-liner from TNG.
 
1. DS9 - thanks to the Ferengi and some memorable Weyoun and Dukat scenes.
2. TNG - mostly because of Q and sometimes Data being involved
3. TOS - Spock vs McCoy and Trelane, not to mention the Tribbles
 
This question can only be answered by asking which series proved that Iggy Pop as an undead Vorta was not all that dissimilar from Iggy Pop.

They could have put him into a Jem'Hadar costume, but that wouldn't have been as funny as Iggy as a Vorta.
 
Deep Space Nine. First to even have a comic relief character in Quark and not only that he was funny and he was funny despite the fact he was Ferengi who were never all that funny despite them also being used for comic relief on TNG. The underated one though is "Voyager." For all it's flaws the show was actually quite good at humor. The EMH was always a hoot and so was Tom Paris and I don't hate Neelix like so many others seem to do.

Every time a poll is made that ignores The Animated Series, I die a little inside. There are Seven series.
 
My son and I are almost finished with a complete rewatch of Deep Space Nine, and I gotta say, I don't know what you guys are talking about. This is the least funny Trek of all (well, 2nd behind Discovery). Garak and Quark provide some good moments, but otherwise, it's grim.

Voyager was the best at comedy, IMHO. Seven was hilarious (without ever intending to be). The EMH. Tom Paris. Neelix. Even Janeway got in one every now and again ("At ease before you sprain something").

TOS is close behind.

So, I'm in close alignment. DS9 had it's moments...and was successful when it TRIED to be funny, but those moments were few and far between. I'd say the same for DSC (it was funny on the rare occasion it made the attempt)

Do you mean grim as in the attempts at humour or bad, or as in the show's grim thematically?

All of the above. The show is grim, and the few attempts at humor (outside Garak and Quark) misfire, IMHO. It's very fresh in my mind, as we're just about at the finale.

Discovery is even more grim. But Tilly gets a laugh here and there.

I think on TNG, Data was intended for sole comic relief, before he became a fan and writer favorite and they started treating him seriously. Worf, too, was meant to humorously juxtapose his barbaric Klingon sensibilities with the evolved Human ethos.

But the earliest comic relief character has to be Dr. McCoy. Man Trap and FTWIHAIHTTS aside, almost every episode, his contributions to the plot were mostly jokes.
 
The scene where Worf agrees to temporarily take Data's cat is the single best comedy bit in all of Star Trek.

Spiner and Dorn each get a fair number of funny lines and bits.
