i'd say #1 for me has to be Telsia Murphy from VGR Elite Force and EF2

I don't really think of her as a "babe", but Alyx Vance. Half-Life 2 and the episodes did such a good job of making the player feel like her and Gordon were a team. She's my girl, she's always got my back.

No other game has managed to make a NPC feel like you can rely on them like HL2 has.

Tifa (Final Fantasy VII) - not the FF7:AC version
Aeris (Final Fantasy VII) - not the FF7:AC version
Nova (the illfated Starcraft Ghost) - the character model had the hall marks of a Lara Croft beater, especially considering the rather large amount of fanart the character has even though the game hasn't been released.
 
I second both Tifa and Aeris from Final Fantasy VII - I loved how Aeris looked in Kingdom Hearts 1 too.
