I'll lead off with the easy one:

There are a lot of actors with two successful franchises and/or TV series to their name, but who are lacking a third, sometimes because they turned down sequels to some of their hits or those sequels didn't come together.

For instance, Schwarzenegger has two - Conan and Terminator - and would have three, or more, if he'd made sequels to Predator, Total Recall, or True Lies (he turned down the first, while sequels to the other two were talked about but didn't come together).
 
If the Sin City franchise ever gets under way:

Patrick Stewart has two big ones: Professor X and Jean-Luc Picard. He needs a third.
Karla from Tinker, Tailor, Soldier, Spy / Smiley's People?

Of course, some actors have several roles they can pick from. For example:

Jim Robinson (Neighbours) - his most famous role
Caleb Nichol (The OC)
Bradford Meade (Ugly Betty)

to name but three.
