The best quote you ever heard, from any character.

It can be any of the shows or movies.

Lets try to exclude the original star trek and enterprise though... the original series was before my time and enterprise was just terrible.

It can be from any shows except the ones you don't like.

Mmh, the quotes that immediately came to my mind when I read the thread title are from TOS or ENT, funnily enough.

    TNG: Q Who?

Q: If you can't take a bloody nose, go home and crawl under your bed. It's not safe out here. It's wondrous, with treasures to satiate desires both subtle and gross. But it's not for the timid.

I'm going to ignore the shot at Enterprise.

    Terra Prime:

Captain Archer: Up until about a hundred years ago, there was one question that burned in every human, that made us study the stars and dream of traveling to them, Are we alone? Our generation is privileged to know the answer to that question. We are all explorers, driven to know what's over the horizon, what's beyond our own shores. And yet, the more I've experienced, the more I've learned that no matter how far we travel, or how fast we get there, the most profound discoveries are not necessarily beyond that next star. They're within us, woven into the threads that bind us, all of us, to each other. The final frontier begins in this hall. Let's explore it together.

That would have been my choice as well. I always liked this one by Spock, too:
"After a time, you may find that having is not so pleasing a thing, after all, as wanting. It is not logical, but it is often true."

KIRK: Dammit, Bones, you're a doctor. You know that pain and guilt can't be taken away with the wave of a magic wand. They're things we carry with us, the things that make us who we are. ...If we lose them, we lose ourselves. I don't want my pain taken away. I need my pain.

Maybe not "the best", but certainly one of my favorites.
