How long did the Defiant hold out against the Borg?
Depends when they joined the battle, the Defiant was designed from the keel up to fight the Borg.

How'd that work out for it?

Pretty good actually. I checked the transcript, the Defiant was in the battle from the start, it survive in comabt (however long that was) all the way to Earth. Data reported that there was "heavy damage" to the Borg cubes outer sections, if the Defiant (and other ships) hadn't created this damage, would Picard's targeting the remaining fleet's combine firepower on a select spot within the cube have been possible?

The Defiant work just fine.

I also thought the battle agains the Borg cube took at least a day.

But Picard orders red alert before they warp. It seems a bit of a stretch to do that days or weeks in advance of the battle
Red alert means for the crew to stay alert. That means a difference in shifts, more training, work on the tactical systems, blablabla. Explained!

Isn't that what Yellow Alert is for?

I remember the novel just mentions that the battle starts in the Typhoon sector and then moves onto Earth with the fleet following the cube. It doesn't mention a time span though.

How many sectors were there between the Typhon Sector and Sol Sector? Sectors were said to be about 20 light-years across, if a ship like the Enterprise were able to maintain maximum warp, it could possibly cross that distance in half a day or so.
