Excelsior-class for me, i've always felt that class looks the most elegant. Plus I prefer the blue/green computer terminals of that era over the orange/yellow of the tng-era.
In a TNG context that would be Andrew Probert's Ambassador Class for me. It features the best elements of both worlds, i.e. Excelsior and Galaxy Class. Other than that it's Kirk's television Enterprise. It's a truly timeless design. It looked outrageously futuristic back in the 1960's and still does so today. Bob
I'd pick the Sovereign class. Because it has the best chance to get into and finish the battle. It's the best armed and defended. And has performance capabilities second to none. It has the resources to handle the largest range of mission profiles. And I'd like to see John Eaves' shuttlebay with all of the Argo-class shuttles and their mission packages. And most importantly; No Frakking Kids On My Ship!

Danube-Class Runabout. Nice and compact, easy to park. Probably gets good Light Years per Antimatter.
Hmm... from canon? Galaxy, easily. Best-looking, well-rounded, interesting missions. I want to say Akira, too, but... we really don't know anything about that damned class. But, if we go with the interpretation that makes it a combat-focused cruiser, not necessarily to the extent of the Defiant but more so than most ships its size (so it could still engage in some exploration, but would always be expected to be ready to respond to a tactical situation), then I'd be up for that, as well. And speaking of non-canon things: if we are including entirely fan-made ships, this would be one of my top choices as well. I absolutely love the design, and back when I had my own ideas for Trek fics, I saw this and immediately envisioned it as a cutting-edge "light explorer" (i.e. more or less what Voyager seemed to be) launched in the late 24th century, which could also serve as a new tech testbed.
I'd convert one of the mammoth cargo bays of my Vengeance-class Dreadnought to a huge jungle-like arboretum which I would fill with lions. And a pool. With sharks. With frickin' laser beams.
Defiant would be my first choice. What's not to like? It has ablative armor, loaded quantum torpedoes and ready to take on the galaxy. And, if I decide to go rogue, who could stop me? My second, non-canon, choice would be this baby... for many of the same reasons. Thanks, Saito S, for sharing this one.
